| I. Background | |
| Programmed Cell Death Protein 1（PD-1） ，一種在激活的 T 細胞上表達的受體，與其 配體 PD-L1 和 PD-L2 結合，負向調節免疫反應。 PD-1 配體存在于大多數癌癥中，PD- 1:PD-L1/2 相互作用會(huì )抑制 T 細胞活性，并使癌細胞逃避免疫監視。 PD1/PDL1 信號轉導通 路是腫瘤免疫抑制的重要組成部分，可以抑制 T 淋巴細胞的興奮，增強腫瘤細胞的免疫耐 受，從而實(shí)現腫瘤免疫逃逸。PD1 與 PDL1 結合可以減弱 T 細胞介導的免疫監視，導致免 疫反應缺失，甚至導致 T 細胞凋亡。PD1/PDL1 抑制劑可解除抗腫瘤 T 細胞的免疫抑制， 從而導致 T 細胞增殖并滲透到腫瘤微環(huán)境中并誘導抗腫瘤反應。PD-1:PD-L1/2 通路還參與 調節自身免疫反應，使這些蛋白質(zhì)成為多種癌癥以及多發(fā)性硬化癥、關(guān)節炎、狼瘡和 I 型 糖尿病的有希望的治療靶點(diǎn)。 | |
