| I. Background | |

EGFR（Epidermal Growth Factor Receptor）是上皮生長(cháng)因子（EGF）細胞增殖和信號 傳導的受體，EGFR 屬于 HER 受體家族，該家族由四種相關(guān)蛋白 EGFR（HER1/ErbB1）、 ERBB2（HER2）、ERBB3（HER3）和 ERBB4（HER4）組成，屬于酪氨酸激酶型受體， 位于細胞膜表面，靠與配體結合來(lái)激活。EGFR 與腫瘤細胞的增殖、血管生成、腫瘤侵襲、 轉移及細胞凋亡的抑制有關(guān)。
HER 受體通過(guò)與不同的配體結合而被激活，包括 EGF、TGFA、肝素結合 EGF 樣生長(cháng) 因子、雙調蛋白、β細胞蛋白和上皮調節蛋白。配體與受體的細胞外結構域結合后，受體形 成功能活性二聚體[EGFR-EGFR（同型二聚體）或 EGFR-HER2、EGFR-HER3、EGFR- HER4（異二聚體）]。二聚化誘導酪氨酸激酶結構域的激活，從而導致多個(gè)酪氨酸殘基上 的受體自磷酸化。從而導致一系列接頭蛋白（如 SHC、GRB2）的募集，并激活一系列細 胞內信號級聯(lián)反應以影響基因轉錄，進(jìn)而導致癌細胞增殖、減少細胞凋亡、侵襲和轉移， 并刺激腫瘤誘導的血管生成。 |
